چکیده

A new antibiotic, leucinostatin, was isolated from the culture filtrate of Fenicillium lilacinum. The antibiotic was obtained in the form of white prisms, decomposing at 131~136°C, and showing optical rotation [^]d2+644° (c 0.5, methanol). The ultraviolet absorption spectrum showed only an inflection at 220nm in methanol. The antibiotic gave a positive reaction to biuret and negative to ninhydrin. Upon hydrolysis, it gave a positive reaction to ninhydrin, yielded leucine predominantly, and the peptide nature of the antibiotic was assumed. Leucinostatin was found to be active against some gram-positive bacteria and a wide range of fungi. It was also cytotoxic to HeLa cell culture and showed some inhibitory effect on Ehrlich subcutaneous solid tumor. The LD50 by intraperitoneal injection in mice was calculated to be 1.6mg/kg. The antibiotic manifested a significant hypotensive effect on rabbit blood pressure without affecting epinephrine and acetylcholine responses.

Biosynthesis of Antibiotic Leucinostatins in Bio-control Fungus Purpureocillium lilacinum and Their Inhibition on Phytophthora Revealed by Genome Mining

Purpureocillium lilacinum of Ophiocordycipitaceae is one of the most promising and commercialized agents for controlling plant parasitic nematodes, as well as other insects and plant pathogens. However, how the fungus functions at the molecular level remains unknown. Here, we sequenced two isolates (PLBJ-1 and PLFJ-1) of P. lilacinum from different places Beijing and Fujian. Genomic analysis sh...

First Report on Isolation of Penicillium adametzioides and Purpureocillium lilacinum from Decayed Fruit of Cheongsoo Grapes in Korea

Two species, Penicillium adametzioides and Purpureocillium lilacinum, were isolated from decayed grapes (cv. Cheongsoo) in Korea. Each species was initially identified by phylogenetic analysis of a combined dataset of two genes. Internal transcribed spacer (ITS) and β-tubulin (BT2) genes were used for identification of Penicillium adametzioides, and ITS and partial translation elongation factor...

Studies on peptide antibiotics, leucinostatins. I. Separation, physico-chemical properties and biological activities of leucinostatins A and B.

Leucinostatin, a peptide antibiotic, was separated by silica gel and alumina column chromatography into two related components designated as leucinostatin A hydrochloride (C61H111-N11O13.HCl) and leucinostatin B hydrochloride (C61H109N11O13.HCl). Physico-chemical as well as biological properties of the two separated components were analyzed. These properties pointed to closely resembling chemic...

Structure activity studies on chemically modified homologues of the antibiotic phytotoxic leucinostatin A.

The synthesis and a conformational study of a number of homologues of the well known antibiotic, phytotoxic leucinostatin A are reported. The circular dichroism of all the compounds are discussed. Some conclusions on the SAR of these compounds are drawn. The influence of the alpha-helical conformation and/or the increased lipophile character on their interesting biological activities is emphasi...

The nonapeptide leucinostatin A acts as a weak ionophore and as an immunosuppressant on T lymphocytes.

Earlier studies have disclosed that leucinostatin A, a hydrophobic nonapeptide antibiotic, assumes an alpha-helical secondary structure in nonpolar environments. The present report demonstrates that the peptide acts as a weak ionophore facilitating the transport of mono-and divalent cations through the plasma membrane of T lymphocytes and through artificial membranes. Leucinostatin A does not c...
